Zachariah: "You're bleeding."
Dean [looks down]: "Oh yeah, little insurance policy in case you dicks showed up." [Dean slams his palm onto a angel-banishing sigil] "I learned that from my friend Cas, you son of a bitch." 5.01
[Cas touches their chests, the brothers flinch and groan.]
Dean: "What the hell was that?"
Cas: "An Enochian sigil. It'll hide you from every angel in creation. Including Lucifer."
Dean: "What, did you just brand us with it?"
Cas: "No, I carved it into your ribs." 5.01
"I've killed two angels this week. Those are my brothers. I'm hunted, I rebelled and I did it, all of it, for you." Cas to Dean 5.02
"When humans want something really really bad... we lie." Dean to Cas 5.03
Dean: "Cas, we've talked about this. Personal space?"
Cas: "My apologies." 5.03
Dean: "Give me one good reason why I should do this."
Cas: "I need your help, because you are the only one who'll help me. Please." 5.03
"So what, I'm Thelma and you're Louise and we're just gonna hold hands and sail off this cliff together?" Dean to Cas 5.03
Dean: "Do we have any chance of surviving this?"
Cas: "You do." 5.03
"Well, last night on earth. What... What are your plans? (...) There are two things that I know for certain: One, Bert and Ernie are gay. Two, you are not gonna die a virgin. Not on my watch." Dean to Cas 5.03
Dean uses his self-proclaimed 'best line' on Cas. (Anna: "You know, our last night on earth. All that." Dean: "You're stealing my best line." 4.10)
"It's been a long time since I've laughed that hard. Oh, it's been more than a long time. Years." Dean to Cas 5.03
The way Cas inclines his head to look at Dean in that scene, he's like: 'Dean, why are you bent over like that? Are you hurt? Why are you laughing if you're hurt?' Cas just has no idea what's going on. He has no idea, why Dean is laughing and he's just switching between looking around and watching Dean, so utterly confused bending over to try and figure it. But in the end he's like: 'Dean, I don't know why you are laughing, but I really like it and it fills me with happiness.' Cas really is clueless, but he's smiling because Dean happy makes Cas happy.
Dean: "Hey, you okay?"
[Cas looks straight ahead, stays silent]
Dean: "Look, I'll be the first to tell you that this little crusade of yours is nuts, but, I do know a little something about missing fathers."
Cas: "What do you mean?"
Dean: "I mean, there were times when I was looking for my Dad when... all logic said that he was dead. But I knew, in my heart, that he was still alive. So who cares what some Ninja Turtle says? Cas, what do you believe?"
Cas: "I believe he's out there."
Dean: "Good. Then go find him."
Cas [looks at Dean]: "What about you?"
Dean: "What about me? I don't know. Honestly? I'm good. I can't believe I'm saying that, but, I am, I'm, I'm really good."
Cas: "Even without your brother?"
Dean [bitter]: "Especially without my brother. I mean I spent so much time worrying about the son of a bitch... I mean, I've had more fun with you in the past twenty-four hours than I've had with Sam in years." [smiles] "And you're not that much fun. Funny, you know, I've been so chained by my family, but now that I'm alone... hell, I'm happy." [Dean looks over; the shotgun seat is empty. His smile falters.] 5.03
Dean: "I've been there. Big expert on deadbeat dads, so... And I get it, I know how you feel."
Cas: "How do you manage it?"
Dean [swallows]: "On a good day you get to kill something."
Cas on the phone: "Where are you now?"
Dean: "Kansas City." [Dean leans across the bed to grab his room key off the bedside table] "Century Hotel, room 113."
Cas: "I'll be there immediately."
Dean: "Whoa whoa whoa, no, no, come on man, I just drove like sixteen hours straight, okay? I'm human. There's stuff I gotta do."
Cas: "What stuff?"
Dean: "Eat, for example; in this case sleep, I just need like four hours every once in a while, okay?"
Cas: "Yes."
Dean: "Okay, so you can pop in tomorrow morning."
Cas: "Yes. I'll just..." [Dean hangs up, Cas hears a dial tone] "... wait here, then." 5.04
Castiel spends a whole night on the street waiting for Dean, although he knows that it will be hours before he has slept enough. Cas could go anywhere, he's an angel, but he decides to wait for Dean.
(Dean: "What do you normally do when I'm gone?"
Cas: "Wait for you to get back.")
Dean: "That's pretty nice timing, Cas."
Cas: "We had an appointment."
Dean puts a hand on Cas' shoulder: "Don't ever change." 5.04
Endverse!Cas: "I thought you'd gotten over trying to label me."
Dean: "Cas, we got to talk."
Endverse!Cas: "Whoa... strange. You... are not you. Not now you, anyway. What year are you from?" 5.04
It doesn't matter what timeline or universe, whether he's human or angel or even stoned out of hin freaking mind... Cas knows Dean.
Endverse!Cas: "I wish I could just, uh, strap on my wings, but I'm sorry, no dice."
Dean: "What happened to you?"
Endverse!Cas: "Life." 5.04
(You. You happened to me.)
"What? I like past you." Endverse!Cas to Endverse!Dean 5.04
"I'm not gonna lie to you. Me and him. It's a pretty messed-up situation we got going. But believe me, when you need to know something, you will know it. Until then, we all have work to do." Endverse!Dean to Dean while he's pointing to Endverse!Cas 5.04
Dean turns around because Endverse!Dean wasn't pointing at him. (The camera even goes from blurry to clear and they show a Dean reaction shot.) That is the exact moment when Dean found out that he and Cas in the future had a sexual relationship. He probably had high suspicions when he saw Endverse!Cas, but that and the conversation that follows seal it for him.
Dean: "Hey Chuck, is Cas still here?"
Endverse!Chuck: "Yeah, I don't think Cas is going anywhere." 5.04
The first thing that Dean asks Endverse!Chuck about. Chuck looks thoughtful and then he's smiling at Dean, seemingly unsurprised and almost a little teasing. Almost a little knowing.
Why isn't Chuck surprised at the question? Chuck thinks that this is Endverse!Dean, and Endverse!Dean should know that Cas is here. So why does Chuck phrase it this particular way? Why doesn't he say: "Of course Cas is here, why are you asking? Why are you acting suspiciously? Are you infected?"
Chuck just witnessed an unpleasant scene between Risa and Dean. Dean expresses regret and immediately switches the conversation to Cas. And Chuck is not surprised that Dean's mind goes from sleeping with Jane and sleeping with Risa... to Cas. That Dean links a love interest and sexual partner with Cas. Chuck reassures Dean that Cas is still here, will be still here, no matter what. Cas hasn't gone anywhere, even though he could have. Chuck knows this because he knows that Cas and Dean are lovers, just like Risa knows, like Jane knows. Cas is the one Dean has had a long term relationship with, and Cas is the one that Dean feels the need to talk to now.

When Dean decides he's ready to die, Cas decides that he's ready as well.
"You mean you're gonna feed your friends into a meat grinder? Cas too? Oh man, something is broken in you." Dean to Endverse!Dean 5.04
So Cas doesn't seem to be 'just a friend'... Dean knew that Cas was and has always been more than a friend and he for sure knew that Cas was more than a friend to Endverse!Dean.
"The thing that makes Dr. Sexy sexy is the fact that he wears cowboy boots. Not tennis shoes." Dean 5.08
Dr. Sexy is a fictional character which is a guilty pleasure of Dean's. Dean denied having a crush on him until Gabriel sends Sam and Dean into a realm in which Dr. Sexy is real.
"Okay, Huggy Bear, just don't lose him." Dean on the phone to Cas 5.10
Do you remember Dean's t-shirt with the teddy print and the inscription 'I wuv hugz'?
Dean spots Cas collapsed against the nearest car, his nose bleeding.
Dean: "Hey! Are you all right?"
Cas: "I'm fine. I'm much better than I expected."
Sam and Dean try to help Cas up, but he spits blood and passes out. 5.13
Sam: "If Cas doesn't recover... Is he alright?"
Dean: "What do I look like Dr. Angel, medicine woman? He'll wake up. He's, you know, tough for a little nerdy dude with wings." 5.13
Anna: "I can't find you."
Dean: "Oh, Cas did this thing." [lays a hand on his heart]
Anna [looks away]: "Cas. Right." 5.13
You can literally see the moment Anna realized: Shit, Cas is totally replacing me here. That awkward moment when you realize your brother is hooking up with your ex-boyfriend.
"Oh Cas, not for nothing, but the last person who looked at me like that... I got laid." Dean to Cas
But Cas just gives him the gaze harder. When girls flirt with him, he's an awkward little baby and doesn't know how to respond, but when Dean flirts with him he's suddenly in full top mode.
Cas: "What I don't understand is... where is your hunger, Dean? Well, slowly but surely, everyone in this town is falling prey to Famine, but so far, you seem unaffected."
Dean: "Hey, when I want to drink, I drink. When I want sex, I go get it. Same goes for a sandwich or a fight."
Cas: "So... you're saying you're just well-adjusted?"
Dean: "God, no. I'm just well-fed." (...)
Dean: "You want to go over the plan again? Hey, Happy Meal. The plan?"
Cas: "I take the knife, I go in, I cut off the ring hand of Famine, and I meet you back here in the parking lot."
Dean: "Well, that sounds foolproof." (Castiel disappears) "This is taking too long." (Dean gets out of the car, in Biggerson's Restaurant) Dean: "Cas! Cas..." (Castiel is on the floor. Two demons attack Dean, and bring him to Famine.)
Famine: "The other Mr. Winchester."
Dean (gesturing at Castiel): "What did you do to him?"
Famine: "You sicced your dog on me. I just threw him a steak. Doesn't take much-- hardly a push. And yet, you're all still starving because hunger doesn't just come from the body, it also comes from the soul."
Dean: "It's funny, it doesn't seem to be coming from mine."
Famine: "Yes. I noticed that. Have you wondered why that is? How you could even walk in my presence?"
Dean: "Well, I like to think it's because of my strength of character."
Famine: "I disagree. Yes. I see. That's one deep, dark nothing you got there, Dean. Can't fill it, can you? Not with food or drink. Not even with sex. Oh, you can smirk and joke and lie to your brother, lie to yourself, but not to me! I can see inside you, Dean. I can see how broken you are, how defeated. You can't win, and you know it. But you just keep fighting. Just... keep going through the motions. You're not hungry, Dean, because inside, you're already... dead." 5.14
(Castiel and Dean stand outside Bobby's panic room door. In the panic room - Sam screams)
Sam: "Let me out of here, please! Help!"
Cas: "That's not him in there. Not really."
Dean: "I know."
Cas: "Dean, Sam just has to get it out of his system. Then he'll be--"
Dean: "Listen, I just, uh... I just need to get some air." (outside) "Please... I can't... I need some help. Please?" 5.14
"You know what? Blow me, Cas." Dean to Cas 5.18
In this episode Dean is pushing everyone away, trying to make them want him to say 'yes' so he'll be gone. He tells Bobby "You're not my father" knowing that's where it'll hurt him the most. He tells Sam "I don't believe in you" knowing that will cut the deepest. But what does he say to Cas? He makes a bunch of mocking sexual references. He knows that Cas is attracted to him, and this is him calling him out on it to try and make Cas angry and hurt. This episode is Dean trying to convince his family that they don't want him, so he goes for their weak spots. And Cas weak spot is the fact that he's in love with Dean.
"I gave everything for you!" Cas to Dean 5.18
Sam: "What the hell happened to him?"
Cas: "Me." 5.18
Dean: "Whoa, wait. You're gonna take on five angels?"
Cas: "Yes."
Dean: "Isn't that suicide?"
Cas: "Maybe it is. But then I won't have to watch you fail." 5.18
Dean on the phone: "Cas? We all thought you were dead. Where the hell are you, man?"
Cas: "A hospital."
Dean: "Are you okay?"
Cas: "No."
Dean: "You want to elaborate?"
Cas: "I just woke up here. The doctors were fairly surprised. They thought I was brain-dead."
Dean: "S-so, a hospital?"
Cas: "Apparently, after Van Nuys, I suddenly appeared, bloody and unconscious, on a shrimping boat off Delacroix. I'm told it upset the sailors. (...) I can't 'zap' anywhere. You could say my batteries are... are drained. I'm saying that I am thirsty and my head aches. I'm saying that I'm just incredibly..."
Dean: "Human. Wow. Sorry."
Cas: "Well, my point is... I can't go anywhere without... an airplane ride. And food. And more pain medication, ideally."
Dean: "All right. Well, look, no worries. We'll wire you the cash."
Cas: "Dean, wait. You said no to Michael. I owe you an apology."
Dean: "Cas... it, it's okay."
Cas: "You are not the burnt and broken shell of a man that I believed you to be."
Dean: "Thank you. I appreciate that."
Cas: "You're welcome." 5.21
Cas: "If you couldn't reach him here, you're certainly not gonna be able to on the battlefield."
Dean: "Well, if we've already lost, I guess I got nothing to lose, right?"
Cas: "I just want you to understand – the only thing that you're gonna see out there is Michael killing your brother."
Dean: "Well, then I ain't gonna let him die alone." 5.22
And of course Cas goes with him.
Dean: "Cas, you're alive?"
Cas: "I'm better than that." (heals him)
Dean: "Cas, are you God?"
Cas: "That's a nice compliment. But no. Although, I do believe he brought me back. New and improved." 5.22
